I have one pardini cylinder, 250 bar can it be filled by the cycle shops ( they have big cylenders to fill cycles) or petrol pump car inflator?

Short answer - No you cannot.
Long answer:-
250 BAR = 3,600 PSI (approx)
Tyre inflators are typically rated to 40 PSI , you can maybe find some rated somewhat higher, but NONE of them will be rated for thousands of PSI.
